Primary liver cancer, or liver cancer for short, causes about 700,000 deaths worldwide each year, and more than half of the deaths come from China.

July 12, 2021 Peking University Biomedical Frontier Innovation Center (BIOPIC), Beijing Future Gene Diagnosis Advanced Innovation Center (ICG), Researcher Bai Fan, School of Life Sciences, Professor Zhu Jiye's team of Hepatobiliary Surgery, Peking University People’s Hospital, and Peking University No.

In this study, the researchers conducted a multi-point sampling study on intrahepatic cholangiocarcinoma.

In order to further explore the correlation between ICC heterogeneity and clinical characteristics, the researchers divided patients into a high heterogeneity group and a low heterogeneity group based on the heterogeneity score

On the other hand, through a comprehensive analysis of multiple omics data, the researchers found that the immune microenvironment of the IDH mutant subgroup and the non- IDH mutant subgroup also have greater differences.

Figure 2: IDH mutation subgroup with high heterogeneity and "cold tumor" characteristics

Finally, the researchers conducted in-depth investigations through the single-cell transcriptome sequencing of the four cases, and successfully confirmed two important characteristics of the IDH mutation subgroup at the single-cell level , high tumor heterogeneity and cold tumor microenvironment

The findings of the two characteristics of IDH mutation subgroups in this study have important clinical significance

In summary, this research system clarifies the high heterogeneity of intrahepatic cholangiocarcinoma at the level of genome, transcriptome, and microenvironment, and shows for the first time that the IDH mutation subgroup has high tumor heterogeneity and colder tumor microenvironment.
