    Cancer Cell: Kidney cancer research found T cell characteristics related to immunotherapy response

    A British research team recently discovered some immune cell characteristics that seem to be consistent with the response of renal clear cell carcinoma to checkpoint inhibitor therapy


    Clear cell renal cell carcinoma (ccRcc) is the most common pathological type of kidney cancer, accounting for about 70%-80%, and the global incidence is rising


    In a phase II clinical trial called "Anti-PD-1 (Nivolumab) Therapy as Preoperative and Postoperative Treatment for Metastatic Renal Cell Carcinoma", researchers used exome sequencing and RNA sequencing , T cell receptor analysis and flow cytometry analysis of 115 tumor samples from 15 patients


    The research team analyzed tumor characteristics before and after treatment with the anti-PD-1 compound nivolumab (Opdivo from Bristol-Myers Squibb)


    The researchers pointed out that the therapeutic response also corresponds to enhanced immune infiltration and higher than normal levels of CD8+ T cells, which can enhance the immune-related granzyme B serine protease activity


    They also emphasized the association between ccRcc-specific human endogenous retrovirus (HERV) and cases that did not respond to PD-1 therapy


    "We found that HERV, which is associated with T cell infiltration in large tumor tissue biopsies, is also highly expressed in immune cells," the authors wrote


    In contrast, the researchers did not detect any connection between the treatment response and the molecular characteristics of the tumor itself, such as single-base changes in specific genes, small insertions or deletions, somatic copy number profiles, or tumor mutation burden


    The author points out that this study shows that there are tumor-specific T cells with cytotoxic characteristics in clear cell renal cell carcinoma, which is expected to bring hope to the development of adoptive cell therapy for cancer
