Picture: hsf1-mediated heat shock pathway promotes protein stability of hematopoietic stem cells during culture and aging

As we all know, hematopoietic stem cells (precursors of blood cells) are difficult to culture in petri dishes, and petri dishes are a key tool for basic research

The research results will be published in the Cell Stem Cell online edition on August 12, 2021

In bone marrow transplantation, hematopoietic stem cells are injected into a vein to rebuild blood production in patients with compromised bone marrow or immune systems

"Even for patients receiving stem cell therapy, transplanting more cells will produce fewer complications and increase the chance of overall success," said senior author Robert Signer, PhD, assistant professor of medicine at the University of California, San Diego School of Medicine

When other researchers tried to cultivate hematopoietic stem cells by rebuilding the bone marrow environment in a petri dish, Signer's team turned to ask what internal mechanism made the cells unhealthy in the first place.

The lead author of the study, Miriama Kruta, a postdoctoral and Ph.

Signer said: "Now, we can store high-quality stem cells in culture for a long time

Both mouse and human hematopoietic stem cells show that enhanced HSF1 activation can restore protein homeostasis

Researchers have discovered that the heat shock pathway is not only important in a petri dish, it can also keep the stem cells in the native bone marrow healthy during aging

Signer said: "HSF1 is activated during aging to maintain the health of stem cells
