Stem cell transplantation does not cause changes in the DNA of the donor cells

Stem cell transplantation is an important part of treating some children with leukemia

    The safety of transplantation

Researchers from Van Boxtel's team at Princess Maxima Children's Cancer Center studied the entire DNA of blood stem cells from nine patients who had received stem cell transplants

They found that most transplanted blood stem cells did not contain more mutations than healthy donors

The research was published in the journal Cell Stem Cell and was supported by funders such as NWO and the European Research Council

    Unknown "scar" in DNA

In a small number of patients, the Van Boxtel team, also affiliated with the Oncode Institute, discovered specific DNA mutation patterns or "scars" that they had never seen before

In this way, the scientists found nine more patients with this DNA scar in their cancer

    Cancer gene mutation

To study whether ganciclovir actually caused DNA changes, the research team exposed cultured blood stem cells to ganciclovir in the laboratory

The team then tested another antiviral drug: foscarnet, which is also used to treat viral infections after stem cell transplantation, but due to the side effects on the kidneys, most patients cannot tolerate it for a long time

In order to better understand the effects of ganciclovir on DNA, longer-term further studies are now needed in a larger patient population

"Thanks to new technologies that uncover the entire genetic makeup of tumors, we have discovered that an antiviral drug may have carcinogenic effects

"The exact effect of ganciclovir on the scars left by the DNA is still unclear

"There are currently more than 30 antiviral drugs of this type on the market that are used to combat various viral infections

The author said, "It is gratifying that stem cell transplantation is a method of treating children and adults with leukemia and other blood diseases.

"Thanks to this research, we now know that ganciclovir treatment after stem cell transplantation may have long-term side effects in some patients

"Viral infections after stem cell transplantation can be serious and sometimes life-threatening
