    In recent years, the application of the key genes Bbm and Wus2 in plant stem cell development in improving plant transformation efficiency has been reported in foreign countries, confirming that regulating the spatiotemporal expression of Bbm and Wus2 can effectively improve the genetic transformation efficiency of important crops such as mai.


    Chinese researchers first tested the effect of the introduction of Bbm-Wus2 expression cassette assisted transformation on the transformation efficiency of maize backbone inbred lin.


    At the same time, the target gene vector and the Bbm-Wus2 expression cassette vector were mixed and transfected, and it was found that plants that did not contain the Bbm-Wus2 expression cassette and were positive for the target gene could be obtained in the T0 generation, but the separation rate of high-quality transformed seedlings (positive seedlings did not contain The proportion of Bbm-Wus2 expression cassette) is relatively low (the average value is 22%), which is difficult to meet the high-throughput requirements of industrializati.


    In order to further solve this problem, in this study, a green tissue-specific expression promoter was added to the overexpression helper vectors of Bbm and Wus2 to drive the lethal expression cassette of Bacillus amyloliquefaciens Barnase gene expression, and the mixed transformation technology strategy with the gene editing vector was adopt.


    The study found that most of the callus containing the Bbm-Wus2 expression cassette could not differentiate into seedlin.


    This means that this study successfully established and improved a genotype-independent high-efficiency maize genetic transformation system, which provided strong support for improving biological breeding efficiency and precise improveme.
