    High-throughput analysis of the three-dimensional structure of protein machinery in a complex biological environment

    On September 8, 2021, " The Innovation " magazine published online the research result "Determining structures in a native environment using single-particle cryo-electron microscopy images" of the Zhang Xinzheng group of the Institute of Biophysics, Chinese Academy of Sciences
    .


    This work is a new method of protein identification in superimposed density developed after the block reconstruction algorithm ( Nat Commun 9, 1552) developed in 2018 to achieve structural optimization in the superimposed density of proteins, and the algorithm is extended to In-situ structural analysis


    At present, the structural analysis of cryo-electron microscopy in complex biological environments is mainly based on electron tomography (cryo-ET)
    .


    Cryo-ET needs to tilt the sample stage to collect sequence tilting data from different angles, and the throughput is very low.


    For samples in a complex biological environment, the density of the target protein in the cryo-EM image overlaps with the density of other surrounding proteins.
    Based on this, this research develops a protein recognition algorithm and derives the optimal weight of the detection function.
    The new function improves the efficiency of detecting the target protein in the superimposed density
    .


    Combining the sorting function to reduce the false positive data introduced by the protein recognition algorithm, effectively reduce the impact of model deviation, and achieve high-resolution reconstruction (Figure 1)
