    Now, a team led by Okada has revealed the molecular basis of chronic bone infections in patients with MSMD


    Miyuki Tsumura, a pediatric researcher at the Graduate School of Biomedical and Health Sciences at Hiroshima University and the first author, said: “The frequency of multifocal osteomyelitis in patients with multiple osteomyelitis is particularly high.


    She pointed out that analysis of osteomyelitis lesions showed an increase in the number of osteoclasts, which are responsible for the absorption of old bone cells during the growth and repair process


    The researchers used three osteoclast precursors cultured from bone marrow cells of MSMD patients and healthy volunteers to examine the formation of osteoclasts in the presence or absence of IFN-γ


    When IFN-γ was added to healthy cells, the formation of osteoclasts stopped as expected


    When it is added to the cells of MSMD patients, the response is weakened-these cells resist the call to stop forming


    "These results indicate that in the absence of signaling molecules, IFN-γ-induced inhibition of osteoclast differentiation and bone resorption is impaired, leading to excessive osteoclast proliferation and increased bone resorption at the infection site.


    The researchers plan to further study the overproduction of osteoclasts and the role of osteoblasts (cells that produce new bone) to fully describe the molecular mechanism of multifocal osteomyelitis
