Inoculation technique is the most commonly used basic operation technique in microbiology experiments.
Inoculation is to use inoculation tools to transplant the culture onto a new medium under aseptic conditions

(1) Vaccination tools
Commonly used inoculation tools include inoculation needles, inoculation loops, inoculation shovel, coating rods and so on
.
Inoculation loops or inoculation needles for bacteria or yeast are used.

(2) Common vaccination methods
According to different purposes, inoculation loops, inoculation needles, pipettes, etc.
can be used to inoculate slant medium, liquid medium and semi-solid medium puncture inoculation
.
1.
Slant medium inoculation The method of inoculating microorganisms from one slant medium to another slant medium is called slant medium inoculation
.
Inclined medium inoculation is mainly used to inoculate pure bacteria, which can be used to identify or preserve bacterial species after proliferation.
(1) Hold the strain tube and the culture medium tube in the left hand, with the slope facing upward, and the strain tube on the outside.
Use the right hand to hold the inoculation loop with a writing brush, and sterilize the platinum wire and the handle that is going to enter the test tube by flame
.
(2) The cotton plug of the culture tube is clamped between the little finger and palm of the right hand, and the cotton plug of the strain tube is clamped between the fourth finger and the little finger, and the nozzle of the tube passes through the flame
.
(3) Extend the inoculation loop into the strain tube and dip a small amount of strain
.
(4) The inoculation loop extends into the culture medium tube, and draws a line on the inclined plane, or draws a straight line
.
Be careful not to touch the mouth of the tube and other places with the inoculation loop stained with bacteria
(5) Pass the flame through the nozzle again, and plug the cotton plug into the original test tube
.
(6) Sterilize the inoculation loop

Liquid medium inoculation The liquid medium inoculation method is basically the same as the slope medium inoculation method.
The difference is that when transplanting the picked strains into the liquid medium tube, hold the test tube diagonally and apply the bacteria to the liquid medium.
On the surface of the tube, the strain will be in the liquid after the test tube is upright
.
This method is mostly used in enrichment liquid for enrichment culture, and can also be used for culture before biochemical tests
3.
Semi-solid medium puncture inoculation.
Semi-solid medium inoculation should use an inoculation needle.
The method of using the inoculation needle is basically the same as the inoculation loop
.
During inoculation, the inoculation needle dipped in the bacterial species is pierced down from the surface of the semi-solid medium, but does not touch the bottom of the tube, and the inoculation needle is withdrawn from the original path
