Background: Shenlian Decoction (SL) is composed of Coptis chinensis and ginseng, and berberine and ginsenosides are its main components

Shen Lian Tang (SL) Shen Lian Tang (SL) Coptidis and Panax ginseng both have the effect of mediating glucose metabolism Mediate glucose metabolism

Objective: Intestinal flora disturbance is one of the important factors causing diabetes.

Gut flora disorder Gut flora disorder

METHODS: db/db mice with fasting blood glucose (FBG) higher than 13.

    Fasting blood glucose (FBG)

FBG in the tail vein was measured by a blood glucose meter every 4 weeks for 8 consecutive weeks

Fresh Stool Samples Fresh Stool Samples 16S rRNA V3+V4 Region Sequencing 16S rRNA V3+V4 Region Sequencing Metagenomics Metagenomics

Results: Compared with ginseng and coptis, SL decoction had better hypoglycemic effect and β-cell protection effect

Hypoglycemic and β-cell protection hypoglycemic and β-cell protection

KEGG analysis and metagenomic sequencing showed that SL decoction could reduce the abundance of Prevotellaceae , Rikenellaceae and Helicobacteraceae , which were related to lipopolysaccharide biosynthesis , riboflavin , respectively.

Decreased biosynthesis of lipopolysaccharide by Prevotella, Rikenbacteriaceae, Helicobacteraceae , lipopolysaccharide, riboflavin metabolism , up-regulated peroxisomes, up- regulated abundance of Bacteroidaceae, up- regulated abundance of Bacteroidaceae, promoted starch and sucrose metabolism and pentose gluconate conversion

Conclusion: SL decoction has obvious hypoglycemic effect , and its mechanism may be related to the regulation of intestinal flora by up- regulating acidobacter

hypoglycemic effect hypoglycemic effect regulates intestinal flora by up-regulating acidobacter
