Background: Inflammatory bowel disease (IBD) such asUlcerative colitis (UC) can be relieved by drugs (anti-inflammatory drugs, glucocorticoids and immunosuppressants ), but current drugs for the treatment of inflammatory bowel disease have obvious disadvantages , such as side effects, high cost, recurrence, and even exacerbation
    .

It is necessary to develop new therapeutic strategies from a TCM perspective to manage and treat IBD

Zhenwu Buqi Decoction (ZWBQD) is a traditional Chinese medicine formula modified from Zhenwu Decoction (ZWD), which is composed of Poria, Baishao, Atractylodes, ginger, Codonopsis, Coptis , etc.
It can be used to treat ulcerative colitis
    .

A previous study reported that ZWBQD attenuated dextran sulfate sodium (DSS)-induced colitis in mice

Objective: To understand the systemic effect and mechanism of ZWBQD on experimental colitis in mice
    .

Methods: Mice were randomly divided into 6 groups—control group, DSS group, ZWBQD group with different doses (7, 14, 21 g/kg/d, equivalent to clinical doses of 0.
    71, 1.
    42, 2.
    13 g/kg/d) and Sulfasalazine group (SASP, 50 mg/kg/d)
    .

Chronic colitis was induced with 3 cycles of 1.

The concentrations of TNF-α, IL-1β, IL-17A and IL-6 in colon homogenate were determined by Luminex method
    .

Based on phytochemical analysis results and network databases (TCMSP and TCMID databases), a database of predicted target-signaling pathways was constructed, and the targets were collected and imported into the protein interaction network, and analyzed using the STRING database (http://string-db.

The pharmacological effects and mechanisms of ZWBQD on DSS-induced chronic colitis mice were investigated by RNA sequencing , western blotting and intestinal flora detection

Results: ZWBQD has obvious anti-inflammatory effect by regulating PI3K-AKT, MAPK signaling pathway and NF-κB signaling pathway, and has obvious protective effect on colon damage in DSS-induced colitis mice
    .

In addition, ZWBQD significantly ameliorated DSS-induced gut dysbiosis and abnormal tryptophan catabolism

CONCLUSIONS: This study uses a combination of multi-omics and network pharmacology to investigate the therapeutic effect and mechanism of ZWBQD on DSS-induced chronic colitis in mice
    .

This study found that ZWBQD had significant anti-inflammatory and protective effects against chronic colitis in mice, and identified the main phytochemical components of ZWBQD

Combining Multi-omics and Network Pharmacology Combining Multi-omics and Network Pharmacology

The pharmacological mechanism of ZWBQD action is related to the inhibition of " PI3K-AKT signaling pathway ", " MAPK signaling pathway " and " NF-κB signaling pathway " in colon tissue and regulation of intestinal microbial metabolism
    .

Furthermore, the multi-omics and network pharmacology integrative approach described in this study can be applied in future studies to evaluate the therapeutic effects and pharmacological mechanisms of complementary and alternative medicines, such as traditional Chinese medicine as a new therapy for the treatment of inflammatory bowel disease
