2-amino-1-methyl-6-benzene-methane and s.4,5-b-phenylimidazo (2-Amino-1-methyl-6-phenylimidazo (4,5-b)pyridine, PhIP) is one of the most abundant heterocyclines, and is a common carcinogen in hot-processed protein-rich foods. Studies have shown that PhIP can induce colon tumors in rodents, but its mechanism is unclear. Xiuli Zhao of Tianjin University of Science and Technology's National Key Laboratory for Food Nutrition and Safety and Yan Zhang, Shuo Wang, and Yan Zhang, Shuo Wang, and others from Nanca University School of Medicine aim to explore PhIP-induced colon damage in rats. 16S rRNA gene sequencing and metabolomic results show that PhIP destroys the composition of gut bacteria and affects the metabolism of glycelin phospholipids and liouric acid. At the same time, PhIP inhibits lipid metabolism in the gut bacteria. Notably, transcriptional histology revealed that PhIP significantly inhibited the expression of a collection of genes in the colon associated with steroid hormone biosynthetics, fatty acid extension, fatty acid degradation, and glycoplasmic metabolism pathways. The results provide a new perspective for studying the mechanism of PhIP induced colon damage and provide a theoretical basis for further understanding the toxicity of PhIP.
