South Lake News (Correspondent Zhang Yuting) On March 28, the rape team of Huazhong Agricultural University published a research paper entitled Multi-omics analysis dissects the genetic architecture of seed coat content in Brassica napus in Genome Biology

Rapeseed oil is the third largest edible vegetable oil in the world, and improving the oil content of rapeseed is one of the important goals of rapeseed breeding

Transcriptome-wide association analysis identifies genes significantly associated with husk rate (a 20-day seed development transcriptome-wide association analysis; b 40-day seed development transcriptome-wide association analysis)

The study utilized the genome resequencing data of 382 Brassica napus samples and 257 representative materials selected from them, transcriptome data of two seed developmental stages, through genome-wide association analysis (GWAS) and transcriptome-wide association analysis (TWAS) et al.

Knockout of BnaCCRL reduces the husk rate and lignin content of rapeseed and increases oil content

To verify the gene functions of BnaCCRL and BnaTT8, knockout mutants were constructed using gene editing technology

TWAS significant genes are balanced in carbon source allocation in rapeseed seeds

There was a significant negative correlation between husk percentage and oil content in Brassica napus seeds, and reducing hull percentage was beneficial to seeds accumulating more oil

This study comprehensively analyzed the genetic basis of the husk rate of Brassica napus, and cloned two genes regulating lignin content and oil content

Zhang Yuting, a doctoral student from the State Key Laboratory of Crop Genetic Improvement, Zhang Hui, a master student, and Zhao Hu, a postdoctoral fellow, are the co-first authors.

    【English summary】

    Background

Brassica napus is an important vegetable oil source worldwide.

    Results

Here we provide insights into the genetic basis of natural variation of seed coat content by transcriptome-wide association studies (TWAS) and genome-wide association studies (GWAS) using 382 B.

    Conclusions

This study helps us to better understand the complex machinery of seed coat development and provides a genetic resource for genetic improvement of seed coat content in B.
