(1) xinglinzi mentions that PBS is made with:
NaCl 8.0g, KCl 0.2g, Na2HPO4-H2O 1.56g, KH2PO4 0.2g, pH7.4 sterilization.
I found other literature that used:
NaCl 160g, KCl 4g, Na2HPO4-12H2O 58g, KH2PO4 48g, pH7.4 sterilization.
, that is, the final liquid volume is different, 20 times difference,
but the difference between the two groups is one with Na2HPO4 H2O, the other is Na2HPO4, 12H2O, please ask you whether this is a writing error in the literature or are they useful?
(2) also use the above-mentioned PBS for thinners such as Triton X-100 (1%) and DMSO 100ml (10%)?
(3) are also useful concentrations of Triton X-100 (5%) and DMSO 100ml (10%), what I want to know is (2) and (3) which are commonly used?
: What I do is a single-cell gel electrophoresis of the liver.
1, about PBS
we should use PBS should be right.
the recipe you offer is a 10-times PBS with a 2-litre amount. And some
the amount
reagents is certainly not appropriate. For example, the concentration of potassium ions is a bit large.
2, about Triton X-100 (1%) and DMSO 100ml (10%).
we are equipped with 890mL of lysed liquid, pre-use supplement Triton X-100, 10 ml (1%) and DMSO 100 ml.
3, Triton X-100 (5%) should be fine. But this reagent is not cheap, so when 1% is enough, there is no need for 5%.
the amount of KH2PO4 I knocked up wrong, it should be 4.8g (not 48g), but still out of proportion to your concentration (0.2g)? My question is, is it Na2HPO4 with 1 or 12 crystalline water? For Triton X-100 and DMSO, you mean not diluting direct additions is that the 1% and 10% of the bets are not concentrations but additions, right?
it doesn't matter if I use Na2HPO4, I use one. If you use n water, just make a conversion.
