Recently, the team of researcher Wu Haoxing from the Clinical Magnetic Resonance Research Center of West China Hospital of Sichuan University and the team of Professor Liu Xiaogang of Singapore University of Technology and Design jointly published a paper entitled "Overcoming the Spectral Dependence: A General Strategy for Developing Far-Red and "Near-Infrared Ultra-Fluorogenic Tetrazine Bioorthogonal Probes" research paper reveals the general design method and mechanism of new near-infrared bioorthogonal fluorescent probes, and further expands the application of bioorthogonal chemistry in molecular imaging, in vivo biomedicine and other fields

Fluorescent probes can label biological macromolecules at the subcellular level, reveal their roles in molecular networks, and explore the molecular mechanism of related pathological evolution.

The researchers directly connected the meso-positions of tetrazine and classical dyes such as rhodamine and cyanine through tertiary amines, shortening the distance between tetrazine and the fluorescent nucleus, which is favorable for the strong electronic interaction between the two in space.

Based on the above strategies, the authors synthesized and characterized the meso-aminotetrazine-modified oxy-rhodamine, carbon-rhodamine, silicon-rhodamine and cyanine dye Cy7 fluorescent probes

This kind of probe also has good chemical stability, and the photostability is better than that of some commercial fluorescent probes.
