In this study, Wang Lei’s team found that treatment with DNA methyltransferase inhibitors can significantly prolong the Arabidopsis circadian clock cycle, and that met1-3 mutants with reduced CG methylation and drm1 with non-CG methylation lost The four mutants of drm2 cmt2 cmt3 (ddc2c3) all showed the phenotype of prolonged circadian clock cycle.
In order to explore the regulatory mechanism, the researchers used a joint analysis of transcriptome and Arabidopsis methylome data to identify 7 genes whose transcription levels were increased in met1-3 and ddc2c3 mutants, and the methylation level of the promoter region was significantly decreased.
This research system analyzes the mechanism of SDC through the protein cascade degradation pathway SDC-ZTL-TOC1 mediated DNA methylation to finely regulate the circadian clock cycle, and also provides a theoretical reference for mammals or other eukaryotic DNA methylation to regulate the circadian clock cycle .
