, researchers from the Chinese Academy of Agricultural Sciences, in collaboration with the University of California, San Diego, used RNA as a template for commost recombination repair, and obtained a genetically modified anti-ALS inhibitor-like herbicide-like herbicide rice plant for future generations using nuclear enzyme self-cutting and gene editing systems with RNA/DNA dual cutting capabilities, respectively. For the first time in plants, RNA is used as a template for isoogeneic recombination repair, which opens up a new way of using plant RNA as a template for isotrophysics. The paper was published online March 18 in Nature -Biotech.
, genome editing first produces DNA double-stranded fractures at the targeted location of the genome, which can be repaired by non-iso-end connections or iso-recombination repair pathways. Non-iso-end connections are most commonly used for code-shifting mutations to disrupt gene function, while iso-recombinant repair is mainly used for precise replacement or fixed-point insertion of target sequences. In most species, non-isoid end connections are the most important repair pathway for DNA double-stranded fractures, and the probability of precise repair through isoid recombination repair is particularly low, said
co-author of the paper and a professor at the University of California, San Diego. The same-origin reorganization repair path requires an additional supply of same-origin reorganization as a repair template.Li Shaoya, the first author of the
paper and a doctoral student at the Chinese Academy of Agricultural Sciences, said that the use of molecular biology to evaluate recombination events confirmed the feasibility of using RNA as a template for iso-recombination repair and participating in DNA iso-recombination repair. Unlike the usual DNA templates, RNA templates can be continuously produced in the body through the plant's own transcription system, providing more templates for iso-recombination repair.
Xia Lanqin, author of the paper and a researcher at the Chinese Academy of Agricultural Sciences, told the China Science Journal that further optimizing the efficiency of the strategy on this basis is expected to solve the current problem of low frequency of plant homogeneity recombination, and accelerate the process of accurately improving the important agronomical traits of crops through gene editing techniques, and then directing the breeding process of new species of crops.
