Post-traumatic stress disorder (PTSD) is a mental disorder that occurs in individuals after a major traumatic event, which is caused by a combination of genetics and the environment.
the current research on PTSD genetics, there are high results of heterogeneity, lack of interaction between genes and other urgent problems.
focus on genes that focus on the same biological pathway is an effective way to study gene-gene interactions.
dopamine, as a neurotransmitter, plays an important role in neurobiological processes.
there has been a study of dopamine systems linked to a variety of mental disorders, including PTSD.
therefore, studying the relationship between dopamine gene-gene interaction and PTSD is a possible effective way to reveal the genetic basis of PTSD. Based on this idea
, wang Li Research Group of the Institute of Psychology of the Chinese Academy of Sciences and Zhang Kunlin, an associate researcher at the Center for Genetic and Biomedical Informatics Research, collaborated on the study of three dopamine gene constellations (DRD2/ANNK1, COMT and DBH) in the population after the Wenchuan earthquake in 2008 (156 PTSD cases and 978 healthy population controls, mainly adults).
results show that gene-gene interaction DRD2/ANNK1-COMT (rs1800497 x rs6269) is significantly related to PTSD diagnostic results (P-0.0008055), and the association of rs6269 with PTSD diagnosis is controlled by rs1800497.
this result was validated in another independent Wenchuan earthquake-hit population (P.01329).
Rs1800497 marked the protein density of the dopamine D2 receptor (DRD2), while rs6269 marked the protein level seofa level and enzyme activity of the catanophenol-O-methyl transferase (COMT).
Given that the role of DRD2 is to receive dopamine signals, and COMT's role is to inactivate dopamine, the interaction of the two reveals the effect of the dynamic balance of the dopamine energy system on the occurrence of PTSD.
the study is the first to reveal the effects of dopamine gene-gene interaction on the presence of PTSD, suggesting that dopamine genes can interact to influence the occurrence of PTSD.
this non-linear result is aimed primarily at women and is expected to be extended to the East Asian population.
, the genotype combination of interacting SNPs can provide an important reference for early identification, division and intervention of PTSD.
difference between rs6269 genotype frequencies in case-control samples (A-D) under different rs1800497 genotypes (AA, AG, GG and all genotypes).
showed that the association between rs6269 and PTSD diagnosis was controlled by rs1800497 genotype.
