    Antiretroviral therapy (Art) can effectively improve the number of CD4 + T cells and immune function of HIV / AIDS patients, so as to protect patients from AIDS related diseases.however, it has been found that the incidence of AIDS related non related diseases in HIV / AIDS patients after long-term art treatment is much higher than that in normal people.it is suggested that the immune function of CD4 + T cells or the diversity of TCR still exist after art treatment.at present, a large number of studies have focused on the study of the immunosuppressive status of CD4 + T cells in HIV / AIDS patients.there are few studies on the changes of TCR receptor Library of CD4 + T cells in patients with HIV infection and treatment, especially the evaluation of TCR diversity of CD4 + T cells after long-term art treatment.in the previous studies on TCR receptor Library of T cells from HIV / AIDS patients with ngs, the main research strategy was to analyze the characteristics of HIV antigen-specific TCR CDR3, and there was no description of the overall change of TCR diversity. Recently, he published a research paper on HIV / AIDS in Beijing University V-J pairing instead of CDR3 as a macro indicator, describes the change of CD4 + T cell receptor diversity from damage to recovery after HIV infection to art treatment, and reveals that although long-term art treatment can increase the number of CD4 + T cells in HIV / AIDS patients, its damaged TCR receptor library cannot be fully recovered (Figure below).in the course of the study, the researchers noticed that TCR V-J pairing expression in healthy people had a preference.through data analysis, it is found that TCR V-J pairing preference is highly conservative in healthy individuals and among individuals.the similar V-J pairing preference among different individuals provides a scale to evaluate the overall change of TCR diversity after T cell system is hit.the researchers have established a set of evaluation system including five categories (coverage, top 10% cumulative percentage, diversity, intra individual similarity and inter individual similarity) parameters to evaluate the overall change of TCR receptor library diversity of CD4 + T cells after HIV infection and long-term art treatment ().as expected, most of the V-J pairing parameters of HIV infected patients who did not receive art changed significantly; moreover, after more than five years of long-term art, with the recovery of CD4 + T cell number, most of the V-J pairing preference parameters of CD4 + T cells in patients can also return to the level of healthy people.however, it is surprising that the long-term art treatment still makes the similarity of TCR V-J pairing of CD4 + T cells between patients and healthy people still has significant difference.it is suggested that the types of TCR V-J pairs in patients with restoration of TCR diversity are different.in these long-term art treatments, most (240) of the V-J pairs with different expression were found in the α chain of Na ï vecd4 + T cells reconstructed after HIV attack.and art treatment itself can also lead to changes in the expression of some TCR V-J pairs.among these V-J pairing α chains with different expression levels, those affected by HIV but not recovered by art therapy were mainly concentrated in trav38-2 / dv8, trav8-2, trav8-4, trav38-2 and trav8-4 The expression of trav38-1 and trav12-1 was highly correlated with each other (Fig. a, b).
