Major advances in rice genomics during the last few years have made positional cloning in rice much more efficient. Nipponbare is a model rice genotype being sequenced by the International Rice Genome Sequencing Project Consortium. Here, we describe an efficient procedure of the construction of physical map for positional cloning of resistance gene (
    R
) using the Nipponbare genetic resources. This advanced strategy should be useful for the efficient identification of agronomic important
    R
genes from many resistant rice genotypes, including wild rice species.
