    Many large DNA viruses have multiple copies of homologous repeat regions (hrs), which play a key role in the life cycle of the virus
    .
    In this study, the baculovirus artificial synthesis platform constructed in the early stage was used to delete the genome of Autographa californica nuclear polyhedrosis virus (AcMNPV) at multiple sites at the same time, and artificially synthesized the AcMNPV genome Syn-Δhrs with all hrs removed, and A single hr1, hr2, or hr3 Syn-hr1, Syn-hr2, Syn-hr3 were retained, and the virus rescue was successfully carried out

    Research results show that hrs is not necessary for virus replication, but it can enhance gene transcription in adjacent regions, and the transcription enhancement effect is related to the specific sequence of hrs
    .
    The above research proves the feasibility and superiority of synthetic biology technology in multi-site editing and functional research of large DNA viruses, and also provides new ideas for the optimal design of baculovirus expression vectors
