Three, double-ball pressurized column chromatography
The use of double-connected ball pressurized column chromatography eliminates the need for electricity, and also eliminates potential safety hazards caused by electric sparks or short circuits, and achieves intrinsic safety
Figure 23-4 Schematic diagram (a) and actual operation diagram (b)
The use method of double-linked sphere pressurized column chromatography is basically the same as the above-mentioned nitrogen pressurized column chromatography
Four, vacuum decompression column chromatography
Changing the pressure at the upper end of the column to the pressure at the lower end of the column is a vacuum negative pressure chromatography column, as shown in Figure 23-5
The method of using vacuum decompression column chromatography is as follows:
(1) As shown in Figure 23-5, set up the device
(2) Pour the eluent and connect the pipeline, tie it tightly with a rubber band to fix the connection part, and then open the bottom valve to make the air path unblocked
(3) Adjust the vacuum valve until the eluent flow rate is moderate
(4) When adding eluent, you can pour an appropriate amount of eluent directly from the top, taking care not to aspirate as much as possible
(5) When connecting a bottle of eluent and changing the bottle, first open the tee in the vacuum line (temporarily interrupt the vacuum), remove the full receiving bottle, and then replace it with an empty bottle
Figure 23-5 Schematic diagram (a) and actual operation diagram (b)
(6) Repeat the operations (4) and (5) until the separation ends
Advantages and disadvantages of vacuum decompression column chromatography:
(1) Advantages: The operation is relatively safe, and the main advantage is faster speed.
(2) Disadvantages: The amount of eluent is slightly more than that of pressurized column chromatography
