Recently, Xu Guowang, a researcher in the High Resolution Separation Analysis and Metabolomics Research Group at Dalian Institute of Chemical Physics of the Chinese Academy of Sciences, made new progress in the research of metabolomics deep coverage analysis techniques, and published three consecutive research papers in the journal Analytical Chemistry.
metabolomics is an important tool to study epitotypes, but there are still great bottlenecks in the quality of metabolomics data collection and data analysis, which restrict sized application of metabolomic technology, and the development of metabolomics quantitative detection and identification technology is the development direction of metabolomics technology.
in view of the above bottlenecks, the research team carried out systematic in-depth research, through the development of targeted metabolomics methods, the study of metabolite structure - mass spectrometry characteristic strain, combined structure - chromatography retention law to improve the coverage of metabolite symbiotic analysis, the development of metabolite database to improve the mass structure identification capacity of metabolites.
the team established a new method for the analysis of high-coverage targeted lipid slipids, covering 19 lipids, 3377 lipid ion pairs, covering more than 7,000 lipid molecular structures. the
method has good linearity, repeatability, lower detection lines, higher coverage, better data quality, and is particularly suitable for large-scale lipid histological analysis (Anal Chem.).
the team also established the Liquid Chromatography-High Resolution Mass Spectrometry (LC-HRMS) for glycline analysis, and built a database of 758 species of mycarine, the largest to date.
new strategy for depth coverage analysis based on the LC-HRMS method, hundreds of acetylcarine (Anal Chem.) can be detected in clinical samples at one time.
in addition, the team developed a comprehensive metabolite standard database construction strategy and method, for data collection, data correction, qualitative algorithms, and inter-instrument differences and other aspects of the problem, to propose a system solution, and developed metabolites qualitative database software (Anal Chem.).
the introduction of the above-mentioned new method of metabolomics, it will greatly promote the coverage and large-scale identification of metabolomic analysis.
the research was supported by the National Natural Science Foundation of China project and the National Key Research and Development Program.
