"Alzheimer's disease", or AD for short, is the most common pathological type in senile dementia.

"Alzheimer's disease", or AD for short, is the most common pathological type in senile dementia.

In 2018, there were approximately 50 million people with Alzheimer's disease worldwide, and an elderly person was diagnosed with Alzheimer's every 3 seconds on average .

AD is characterized by progressive neuropathological changes decades before the manifestation of cognitive and clinical decline.

More and more evidence shows that sporadic AD is related to a decrease in amyloid beta (Aβ) clearance.

    Blood vessel

Studies have found that BBB is involved in the removal of solutes (including Aβ) in the brain, which is closely related to the occurrence of AD.

BBB is involved in the removal of solutes (including Aβ) in the brain, which is closely related to the occurrence of AD.

The decrease in water exchange rate across the BBB may be related to the decrease in the body's Aβ clearance rate.

So, is the water exchange rate of BBB really related to the occurrence of AD and the changes in cerebrospinal fluid biomarkers, such as Aβ clearance? To this end, experts from the Department of Neuroscience at the University of Kentucky School of Medicine used special magnetic resonance imaging (MRI) to carry out related research, and the results were published in the latest Alzheimer's Disease journal.

Researchers performed diffusion preparation, arterial spin labeling (DP-ASL) scans of 40 cognitively normal elderly (67-86 years old), which estimated the exchange rate of water on the BBB (kw).

The difference in blood-brain barrier water exchange (kw) between frontal lobe (sorrel), temporal lobe (yellow), parietal lobe (orange) and precuneus (blue)

The results showed that in multiple brain regions including the frontal lobe, temporal lobe, parietal lobe and precuneus, the BBB kw value was positively correlated with the concentration of Aβ42 in the cerebrospinal fluid.

Researchers believe that the low water exchange rate across the BBB is related to the low CSF Aβ42 concentration.
