(i) fungus fall observation
to observe the morphological characteristics and color of fungal bacteria is helpful to the identification of pathogenic fungi. General observation of fungal falles must be the fungus cultured in the sand to protect the weak
legisic
(commonly known as large culture), after several days to ten days of culture before you can see the typical bacteria.
1.
(1) Clinical specimen (suspected fungal infection): hair or dander.
(2) Fungi
species
: newborn cryptic coli, white fake silk
yeast
bacteria, floc-like psoriasis.
(3) sand-protected glucose sloped culture base (containing penicillin, streptomycin).
(4) other: 70% alcohol, sterile physiological saline, flat dishes, etc.
2. Method
(1) Clinical specimens were soaked with 70% alcohol for several minutes, killing surface clutter and washing with sterile physiological saline.
(2) The treated clinical specimens and laboratory-preserved strains were inoculated on 2 to 3 streptomycin-containing sarcosis glucose slope cultures, and several hairs or pieces of dander were inoculated directly on the clinical specimens. Fungal strains are inoculated with sterilized rings by sloped culture.
(3) in a culture box of 22 to 28 degrees C 2 to 3w or 37 degrees C culture 3 to 7d.
(4) observed 1 to 2 times a week, clinical specimens continuously cultured 3w sterile growth can report negative, such as growing bactericidal, should be observed day by day the pattern and color changes of the fall.
3. Results
the fungus fall can be divided into three morphological categories: yeast, yeast-like (type) and silky.
(1) Yeast infrastrucure (observation of new cryptic cultures): the bacteria fall into a round, whiter, with a smooth, moist surface with neat edges, and false mycelium does not reach into the culture base, similar to the staphylococcus precipice.
(2) yeast-like (type) bacteria (observation of white false silk yeast): the surface is similar to the yeast bacteria, but the resulting false mycelium extends into the medium.
(3) silky bacteria: observe a variety of skin silky bacteria slope culture, the surface of the bacteria mostly have gas-like mycelium, the naked eye observed fluffy, powdery, cotton-like, so called silky bacteria, color variety, red hair bacteria purple red, rust-colored wool bacteria rusty or brown, the bottom of the bacteria have nutrients into the culture base.
(ii) the shape of the fungus is
1. The form of mycelium
(1) combed mycelium: mycelium side has most short branches, spacing is not the same, shaped like comb, so the name comby mycelium is commonly found in Xulan hair bacteria.
(2) helicobacteria: Some myceliums are spiral-curved and commonly found in gypsum-like spores and xulan's hair.
(3) racket-like mycelium: puffy at one end of the mycelium section, such as a tennis racket, commonly found in microsporum and epidermophyton.
(4) nodule-like mycelium: mycelium wrapped into nodule-like, commonly found in Oduan spores and so on.
(5) antlers: the end of the silk puffed into antlers, most commonly seen in Xulan hair bacteria.
(6) false mycelium: yeast (e.g. white false silk yeast) mycellular cells do not fall off after prolonging branching, forming a mycelium-like structure.
