Then each tube is added 0.4M trinchloro acetate solution 3.0ml, respectively,filter, each absorb filter 1.0 ml, plus 0.4M sodium carbonate solution 5.0ml, forin reagent 1.0ml, shake well, in 40 degrees C water bath insulation for 15 minutes. After coloring, a 721-type hydrometer is used to determine the light density at a wavelength of 680nm (when compared, with distilled water as a blank) and record it.

1. The activity of salivary amylase varies from person to person and sometimes varies widely, so dilution multiples can vary from person to person.
2. A small amount of agitation or inhibitor can affect the activity of the enzyme. But anthicides and inhibitors are not absolute, and some substances are an enzyme's agitant at low concentrations and inhibitors at high concentrations. One enzyme is an agitant and the other is an inhibitor.
3. Reaction time should be accurate, the utensils used in enzymatic experiments must be clean,dry , quantitatively accurate.
