Original title: Professor Li Zhuoyu: Hulusin E by easing TFAP4/Wnt/β-Catenin signaling pathrax to the colorectal cancer cells chemically increased sensitivity

Therefore, there is an urgent need to identify new advents that make CRC cells sensitive to conventional chemotherapy methods. Cucurbitacin E (CE) is a natural triamcinoid that is widely found in dietary plants and has anti-tumor effects.
this study, Peng Yang, Wen Liu, and Zhuoyu Li, of Shanxi University's Institute of Biotechnology (key laboratory of the Ministry of Chemical Biology and Molecular Engineering), among others, noted that CE enhances the sensitivity of CRC cells to chemotherapy by reducing the expression of box-type transport proteins ABCC1 and MDR1 by weakening adenosine 5'-triphosphate (ATP). Combined with CE's functional magnetite nanoparticles and gene ontogen analysis, it was found that CE binding proteins may involve signaling of the Wnt/β-catenin path. To confirm this finding, β-catenin was raised in drug-resistant cell line, while the synergy between CE and chemotherapy was associated with a reduction in β-catenin expression. In addition, TFAP4 is identified as an in-cell target for CE. It is worth noting that the combination of CE and 5-FU treatment reduced the expression of β-catenin, MDR1 and ABCC1, while TFAP4 over-expression reversed their expression by 2.68±0.46, 0.72±0.44 and 0.93±0.21 times, respectively. Therefore, our results show that CE makes CRC cells sensitive to chemotherapy by reducing signaling of the TFAP4/Wnt/β-catenin pathrapy, indicating that the dietary compound CE can be used as a chemotherapy admissive for CRC therapy.
