    According to the White Paper on Report on the Status of Gout in China in 2021, the number of hyperuricemia patients in China has reached 177 million, of which about 166 million people suffer from gout, and with the improvement of people's living standards, especially the intake of a large amount of high-purine diet , the patients with hyperuricemia showed a clear upward and younger tre.


    In this study, based on the previous stage, leucine was used as the lead compound, and the molecular structure was modified by chemical synthesis, and 30 leucine derivatives were designed and synthesiz.


    This paper is the first to systematically study the uric acid-lowering activity of the synthesized new lotus leaf alkaloid derivatives, which provides strong data support for using it as a lead compound to develop high-efficiency and low-toxicity uric acid-lowering drugs and using lotus leaf to develop healthy functional produc.


    Research Highlights

    The inhibitory effect of compound 1j (1046±07%) in the HK-2 cell model induced by uric acid was approximately the same as that of the positive control drug benzbromarone (1044±21%);

    Compound 1j inhibits the expression of uric acid transporters URAT1 and GLUT9 in HK-2 cells by inhibiting the PI3K/Akt pathway in the cells;

    Compound 1j can promote the excretion of uric acid, thereby increasing the proportion of uric acid excretion in the body;

    Compound 1j can improve renal inflammation and reduce the expression level of inflammatory factors in the kidney;

    Compound 1j exerts renal protection by regulating TLR4/IκBα/NF-κB signaling pathw.
