    Pharmaceutical intermediates, pesticides and fine chemicals occupy an important position in the national economy and people's livelihood


    Diels-Alder reaction (Diels-Alder reaction, referred to as DA reaction) is a [4+2] cycloaddition reaction between conjugated diene (dienoid) and substituted olefin (dienophile), It is one of the most commonly used methods to construct CC bonds in synthetic chemistry.


    On December 10, 2021, Peking University’s School of Chemistry and Molecular Engineering, Peking University-Tsinghua Life Sciences Joint Center Professor Lei Xiaoguang’s research group cooperated with Professor KN Houk’s laboratory at the University of California, Los Angeles, and published an online publication titled “Enzymatic” in Nature Catalysis .


    Inspired by the biosynthesis of DA type natural products in moraceae, the author unearthed and identified two types of FAD-dependent proteins from white mulberry, among which one type of FAD-dependent protein (MaDA-1) has a selective DA reaction enzyme with the known endo MaDA has the same function, and another type of FAD-dependent protein (MaDA-2 and MaDA-3) catalyzes the rare exo-selective intermolecular DA reaction

    The author also analyzed the crystal structure of exo selective DA reaction enzyme MaDA-3, and combined computational simulation and mutation experiments to analyze the mechanism of the two types of DA reaction enzymes with different endo/exo selectivity

    In summary, Lei Xiaoguang's research group and KN Houk's research group have reported two FAD-dependent proteins with different functions, which catalyze the occurrence of different endo/exo selective DA reactions, and confirmed that this type of DA reaction enzyme has a good effect.
