    Yangxuejiedu Decoction (YJS) is a famous traditional Chinese medicine formula for the treatment of " blood dryness syndrome ".

    Objective: To investigate the therapeutic effect of YJS on psoriasis model mice and its molecular mechanism


    Methods: 50 BALB/c male mice were randomly divided into 5 groups (n=10): control group , IMQ group (imiquimod) , YJS+IMQ group , PES (Hsp70 inhibitor)+IMQ group , MTX group (Methotrexate) + IMQ group

    According to the Clinical Psoriasis Area and Severity Index (PASI) , the severity of skin lesions was observed and calculated for 6 consecutive days (days 4-9); skin pathological changes were observed by HE staining ; CD4+ and CD8 were analyzed by immunohistochemistry and immunofluorescence + expression; ELISA was used to detect the levels of inflammatory factors TNF-α, IL-6, and IL-1β in each group; exosomes were extracted using a whole exosome isolation kit , and western blot was used to detect the levels of HSP70, CD63 and Calnexin proteins in exosomes.

    Results: YJS could effectively inhibit the hypertrophy, erythema and scale of psoriatic skin lesions , alleviate the pathological changes of skin lesions, and further reduce TNF-α, IL-6, IL-1β, IFN-γ, IL-17 and IL -23 production

    Conclusion: The therapeutic mechanism of YJS may be achieved by inhibiting the secretion of HSP70 from exosomes , preventing the activation of TLR4, and inhibiting the phosphorylation signaling pathway of NF-κB, further reducing the production of inflammatory cytokines and mediators
