A few days ago, the group standard "Determination of Osteopontin in Infant Food and Dairy High Performance Liquid Chromatography Method" jointly formulated by Heilongjiang Feihe Dairy Co.
    , Ltd.
, National Dairy Science and Technology Innovation Alliance, Zhongyou Dairy Research Institute (Tianjin) Co.
    , Ltd.
, and Beijing Institute of Animal Husbandry and Veterinary Medicine, Chinese Academy of Agricultural Sciences
    , passed the examination.

Osteopontin is a highly phosphorylated bioactive protein
    closely related to immune protection.
The content in human milk is high, which plays an important role
in the immune regulation and intestinal development of infants and young children in early life.
The content of osteopontin in bovine milk is lower than that in human milk, so milk-based infant formula fortified with osteopontin has increasingly become a hot product in the milk-based infant formula market, but the lack of osteopontin detection standards in dairy products in the industry limits the emulsification innovation
    of products.

At present, the main methods for the determination of osteopontin include enzyme-linked immunoassay (ELISA) and liquid chromatography-mass spectrometry
    .
Although the ELISA method is easy to operate, the mainstream method mainly focuses on the determination of human mastopontine protein, and there are fewer antibodies against bovine mastopontine protein, and the accuracy and sensitivity are relatively low
    .
The method of measuring osteopontin by liquid chromatography-mass chromatography has high investment cost, poor accuracy and great interference, especially the Maillard reaction during product processing makes lysine lactosylation modification and pancreatic enzyme action site lose specificity, resulting in a decrease
    in specific peptides.
Studies have shown low levels of osteopontin as measured by mass spectrometry
    .

Based on the above problems, the standard formulation team created a high performance liquid chromatography determination method, selected an ion exchange chromatography column as the separation column, solved the problem of separation and extraction of osteopontin during the pretreatment of different dairy products and infant formula milk powder, and researched and formulated the standard for the detection method of osteopontin "Determination of Osteopontin in Infant Food and Dairy Products High Performance Liquid Chromatography Method
    ".

This method is simple to operate, low input cost and high application popularity, which solves the problem
of missing detection methods of osteopontin in dairy products and infant formula.
