    Plant intracellular immune receptor NLR genes are highly regulated at the transcriptional and post-transcriptional levels to balance plant defense and growth
    .

    At present, the regulation of NLR protein has been deeply studied.
    However, since NLR genes are often located in gene clusters and are regulated by large regions of chromatin, transgenic methods cannot be used to study their cis-elements.
    The regulatory mechanisms of induction, repression or co-expression are poorly understood
    .

    Recently, researcher Zhou Tong's research group from the Institute of Plant Protection, Jiangsu Academy of Agricultural Sciences and Professor Hua Jian's research group from Cornell University published a paper entitled "In situ deletions reveal regulatory components for expression of an intracellular immune receptor gene" on Plant, Cell & Environment.
    and its co-expressed genes in Arabidopsis” research paper, discovered multiple cis factors and two trans transcription factors that regulate the transcriptional regulation of the immune receptor gene SNC1 in plant cells, and initially revealed its regulatory mechanism
    .

    To search for cis-regulatory elements of SNC1, the researchers created a series of deletions at the SNC1 site through a dual-targeted CRISPR/Cas9 system, spanning a region 2 kb upstream of the translation initiation site and 4 kb downstream of the stop codon
    .

    Through the analysis of 10 deletion mutants, multiple transcription-promoting and repressing cis-elements were identified, such as upstream region C2, C5 and downstream region C9 are positive cis-elements, upstream region C8 and downstream region C10 are negative cis-elements type components (Figure 1)
    .

    Transcription factors LHY1 and bHLH28, which may be bound to two transcription promoting elements, were further identified and their functions in the regulation of SNC1 gene expression were verified (Fig.
    2)

    These results provide new clues for the transcriptional regulation mechanism of the plant intracellular immune receptor gene SNC1
