Is there non degradable adsorption However, there is no report on the specific degradation of plant cell wall by cellulase Therefore, effective pretreatment of lignocellulose and visualization of cellulase hydrolysis of plant cell wall will greatly improve the efficiency of lignocellulose biorefinery Recently, researchers from the Biophysics Department of the Institute of modern physics, Chinese Academy of sciences have made new progress in the research on the process of cellulase hydrolysis and alkali treatment of straw The researchers found that when the chemical pretreatment of sweet sorghum straw with dilute alkali, dilute acid and hydrogen peroxide improved the enzymatic hydrolysis efficiency of straw biomass, among which the enzymatic hydrolysis efficiency of dilute alkali pretreatment reached 86.44% In this paper, we use CLSM and real-time microscopy to visualize the hydrolysis process of cellulase, and observe the specific way of cellulase hydrolysis cell wall, that is, cellulase first dissolves the middle of plant cell wall, then moves from the middle to both ends, and finally degrades the corner of cell wall The overall hydrolysis efficiency of Cellulase on cell wall is inverse to the content of lignin in cell wall Ratio The results of CLSM showed that vascular tissue in straw was highly lignified During the hydrolysis of lignocellulose by cellulase, there was non functional adsorption between cellulase and lignin, which reduced the functional adsorption with cellulose Pretreatment with dilute alkali can effectively delignify sweet sorghum straw, reduce the non functional adsorption of lignin and cellulase, and improve the efficiency of enzymatic hydrolysis of plant straw In this work, the pretreatment effect of plant cell wall and the way of cellulase hydrolysis were studied from two aspects: the structural change of straw cellulose and the visualization of enzymatic hydrolysis process The results will help to design plant cell and tissue types from the plant tissue level, so as to reduce its natural anti degradation barrier and improve the conversion and utilization rate of natural lignocellulosic materials Relevant research results are published in biotechnology for biofuels The research was supported by key projects of the Chinese Academy of Sciences (BIOON Com)
