    March 26, 2021 // - Recently, a research report entitled "WNT inhibition creates a BRCA-like state in Wnt-addicted cancer" published in the international journal EMBO Molecular Medicine , from Duke-Singapore Scientists from national universities and other institutions have discovered through research that the anti-cancer drug called ETC-159 may prevent certain tumors from becoming resistant to therapy by blocking a key DNA repair mechanism; ETC-159 is a kind of Anticancer drugs are currently in the early clinical trials of colorectal cancer and gynecological cancer.


    EMBO Molecular Medicine scientists from Duke-National University of Singapore and other institutions have discovered through research that the anti-cancer drug called ETC-159 may block a key DNA repair mechanism to prevent certain tumors from becoming resistant to therapy.

    In many therapies used to treat cancer, poly(ADP ribose) polymerase (PARP) inhibitors can inhibit cancer cells from naturally repairing DNA damage, including unwanted or harmful breaks in DNA.


    Under normal circumstances, the Wnt signaling protein can interact with cell receptors to activate the translocation of a protein called β-catenin into the nucleus, where it can regulate the activation of a variety of genes.


    The researchers pointed out that using a drug called ETC-159 to block the Wnt signaling pathway may reverse the tolerance of many cancer cell lines to PAPR.

    Based on the results of this article, the researchers also found that the same DNA repair signal may help prevent the occurrence of stem cell mutations located in the intestinal epithelium, which further confirms the importance of normal Wnt signaling for stem cell maintenance.
